FOXO4-DRI
FOXO4 D-retro-inverso peptide
Purely a laboratory and animal story so far. The mouse findings are interesting to ageing researchers, but there is no human trial, no human dose, and no way to work out what a human dose would even be from the animal work.
Preclinical senolytic research . Published work is in cells and mice only: there is no established human efficacy, dose, pharmacokinetics or safety.
How well studied
Tier Guide: A = Well studied, B = Some good studies, C = Early evidence & D = Barely studied
Little to no real testing in humans at all. Mostly theory, lab results, or anecdote.
UK status
How you could legitimately come across it here, if at all
No authorised UK medicine contains this, and we have no record of a routine UK clinic route. Material sold online is unregulated.
Evidence strength and UK regulatory status answer different questions.
Heightened caution: this compound carries unresolved regulatory, safety or supply-chain concerns in the UK. Nothing here is a recommendation to use it.
Overview
How it works
In mice and cell cultures, it blocks FOXO4 from holding p53 in the nucleus of senescent cells, which pushes those cells into programmed cell death while sparing normal cells.
D-retro-inverso designed to disrupt the FOXO4-p53 interaction, releasing p53 to trigger apoptosis selectively in senescent cells. Demonstrated in vitro and in murine models only.
Reported benefits
- Preclinical observation: in mice, targeted clearance of senescent cells was associated with improvements in fitness, fur density and renal function markers
- Preclinical hypothesis: selectively removing senescent cells might mitigate features of ageing and of senescence-associated tissue damage
- No human efficacy has been demonstrated for any of this, and none of it should be read as a benefit to a person

Dosage
No established human dose. All catalogued work is preclinical, so there is no human pharmacokinetic or dose-finding data, and no human-equivalent dose can be inferred from the animal experiments.
Side effects
- Human adverse-effect profileunknown; no human studies are catalogued
- Selectivity for senescent cells, and off-target effects on healthy cells in peopleinsufficient human data; mechanism-based concern rather than a demonstrated harm
- Immunogenicity of a synthetic D-retro-inverso peptide in humansinsufficient human data
- Long-term safety of removing senescent cells in peopleinsufficient human data
- Purity and contents of unregulated research-chemical materialunverified
Getting it in the UK
No authorised UK medicine contains this, and we have no record of a routine UK clinic route. Material sold online is unregulated.
Where it turns up
- Not routinely available in the UK
- Unregulated online market
No UK marketing authorisation for FOXO4-DRI was identified, and no authorised medicinal product contains it. Material offered online is unregulated research chemical supply, where nobody is verifying identity, purity or sterility.
No MHRA-licensed product and no identified clinical trial route; material sold online is unregulated
MHRA: the UK regulator that assesses whether a medicine is safe and works before it can be authorised for use here. Something not being MHRA-licensed does not automatically mean it is illegal, and it does not mean it is safe.
This is background information about UK regulation, not advice about what you can or should obtain.

Why this rating?
The A to D rating shown above comes from our earlier editorial review and is still waiting to be migrated into the structured review record below. That is a gap in our record-keeping, not a sign the rating is wrong.
Evidence fingerprint
No route and indication scope has been published for this profile yet, so no fingerprint is shown.
Sources
Structured source audit not yet published for this profile. The references behind this page are being catalogued one compound at a time, and this one has not been through that process yet.
